One of each, DPS Monk, the rest are tower, let the barbarian which remains unused.
All of them! I even have a dps for each of them. Not that they are all good, but they are getting there.

DPS Monk, the rest tower spec(Apprentice, Squire, Huntress, Monk, Summoner, 2x EV(Waller and Buffer))
I have a tower squire, hybrid huntress (will be Traps when I get better gear), tower HP EV, Summoner, Aura Monk, DPS/Boost Monk as my main characters. I am also leveling a Ranger for ranged DPS & an initiate, or whatever the female monk is called, because I wanted to use the name "ButtsLOL" like on my female monk in Diablo 3. It just makes me laugh. So very stupid, but funny to me.
I regularly use a Tower Countess, EV, Initiate, Summoner and Huntress and a DPS Monk (NM) and Barbarian (Insane)
I also have but don't often use a Tower Adept.
I would love to use some of the other characters in combat but in later levels on NM it is too easy to feel like you aren't making a difference without excessive stats.
I have at least 12. In later game play I use the below.

Should probably lvl the hunt and adept to 78 so they can be my builders but haven't really felt like it.
Tower Squire, Tower Monk, Tower Summoner, Tower EV, Hybrid Adept.
Yet technically I guess the Monk and Squire could be considered hybrids too, because each do 1.9-3.5mil damage each hit.. Yay for cats.

Hybrid monk active, tower squire, dps hunt can switch a piece of gear so she can use traps, summoner, ev.
I also have some lowbies to play with friends to run through on insane or medium
